2-Methoxyestrone is an endogenous methoxylated estrogen metabolite formed through catechol-O-methyltransferase (COMT)-mediated O-methylation of 2-hydroxyestrone. As a product of Phase II estrogen detoxification pathways, this metabolite circulates at low levels in human plasma and urine. Alterations in the balance of catechol estrogen hydroxylation and methylation pathways affect cellular estrogen homeostasis, positioning 2-methoxyestrone as an important physiological biomarker for assessing estrogen metabolism and breast cancer risk.
