2-Bromohexadecanoic acid (2-BP) is a nonmetabolisable palmitate analogue acting as a palmitoylation inhibitor that targets palmitoyl-acyl transferases (PATs), inhibiting PAT activity towards MyrGCK(NBD) and FarnCNRas(NBD) with an IC50 of approximately 4 µM. It directly and irreversibly inhibits DHHC family acyltransferases. By blocking protein S-palmitoylation, 2-BP disrupts protein membrane association, subcellular trafficking, and downstream cell signaling pathways. [1][2]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
