Biological Activity

2-Methoxyestradiol is an endogenous estrogen metabolite and potent inhibitor of hypoxia-inducible factor-1α (HIF-1α) activity and microtubule polymerization. By disrupting microtubule dynamics and blocking nuclear accumulation of HIF-1α, it downregulates HIF-mediated transcriptional programs and inhibits angiogenesis. Consequently, 2-Methoxyestradiol suppresses tumor endothelial cell growth and inhibits cell proliferation while inducing both autophagy and apoptosis in various cancer cell types. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
