1,4-Cineole (Isocineole) is a naturally occurring oxygenated monoterpene compound that functions as a dual activator of human transient receptor potential melastatin 8 (TRPM8) and transient receptor potential ankyrin 1 (TRPA1) cation channels. By stimulating these TRP ion channels, 1,4-cineole induces intracellular calcium ion influx and modulates somatosensory neuronal signaling, which downstream regulates cold thermal sensation and neurogenic pain perception mechanisms. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
