Biological Activity
Proteinase K is a broad-spectrum serine endoprotease that cleaves peptide bonds adjacent to the carboxylic group of aliphatic and aromatic amino acids. It is active across a broad pH range of 4 to 12 and retains enzymatic activity in the presence of SDS or urea. Through non-specific proteolytic degradation of nucleases and structural proteins, Proteinase K facilitates nucleic acid purification and assists in isolating resistant amyloid protein cores.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
