    Background
    Phospho-Rb (Ser780) is the phosphorylated, inactive form of the retinoblastoma protein (Rb), a key tumor suppressor and regulator of the G1/S cell cycle checkpoint. Rb contains three major domains—the N-terminal region, a central “pocket” domain critical for E2F transcription factor binding, and the C-terminal region—that together control its interactions with partner proteins. Phosphorylation at Ser780, located within the flexible linker region, is mediated by cyclin-dependent kinases (CDKs) during the G1/S transition. This phosphorylation induces a conformational change in Rb from an extended, active state to a compact, inactive form, thereby disrupting its ability to bind and inhibit E2F. As a result, E2F-driven transcription of genes required for DNA synthesis and cell cycle progression is unleashed, promoting proliferation. Importantly, detection of Rb phosphorylated at Ser780 is used as a functional marker of Rb inactivation, with clinical relevance in cancer prognosis, as loss of tumor-suppressive Rb activity through phosphorylation contributes to malignant progression.
