    Background
Pax6 belongs to the subgroup III paired box transcription factor family characterized by paired and homeodomains that orchestrate tissue-specific gene expression during embryogenesis and lineage maintenance. The protein features an N-terminal paired domain with two helix-turn-helix subdomains for high-affinity binding to consensus TAAT/TCAT motifs alongside a C-terminal homeodomain that cooperates through cooperative DNA contacts and transactivation domains rich in acidic residues. Pax6 maintains radial glial progenitor pools by repressing cell cycle exit genes through direct promoter occupancy at Cdkn1a and Cdkn1b while promoting Wnt3a expression to modulate radial migration via non-canonical planar cell polarity signaling that aligns radial glia scaffolds with cortical layering. Temporal gradients of Pax6 dosage fine-tune G1 phase lengthening through oscillating Cdk6 levels, balancing proliferation with differentiation timing as progenitors transition from symmetric to asymmetric divisions during upper layer neuron generation. Activation occurs via upstream Six3 and Fgf signaling cascades that prime paired domain accessibility, with autoregulatory loops sustaining high rostral expression through chromatin looping to its own enhancer. In pancreatic beta cells driven by Pdx1 and Ngn3 co-expression, Pax6 binds islet-specific enhancers to transactivate insulin biosynthesis via coordination with Pdx1 at the Ins1/2 promoters while regulating glucose-stimulated ATP production through Slc2a2 and Abcc8 control, ensuring Ca2+ dynamics and GLP-1 responsiveness for mature secretory competence.
