    Background
    CD11b (αM, ITGAM) and CD11c (αX, ITGAX) are integrin α-subunits of the β2-integrin family (CD11/CD18), forming heterodimers CD11b/CD18 (Mac-1, CR3) and CD11c/CD18 (p150,95, CR4), respectively, primarily expressed on myeloid leukocytes, including monocytes, macrophages, neutrophils, and dendritic cells. Both feature a large extracellular region with a seven-bladed β-propeller domain, thigh domain, calf-1/calf-2 domains, and an inserted I-domain (von Willebrand factor A-like) as the primary ligand-binding site containing a metal ion-dependent adhesion site (MIDAS) with Mg²⁺ coordination by Asp, Ser, and Glu residues; CD11c/CD18 adopts bent (low-affinity), extended-closed, and extended-open (high-affinity) conformations regulated by talin binding to the β2-tail, with CD11c phosphorylation at α-chain Ser1158 enhancing activation. CD11b/CD18 binds iC3b, ICAM-1, fibrinogen to mediate leukocyte adhesion, transmigration, phagocytosis of opsonized particles, and respiratory burst via PI3K/Akt signaling, while CD11c/CD18 similarly facilitates iC3b/complement-mediated phagocytosis, DC antigen capture (e.g., CD47-deficient cells), and adhesion to endothelium/fibronectin, triggering cytoskeletal remodeling, NF-κB/MAPK activation, and cytokine production for innate/adaptive immunity. Dysregulation contributes to chronic inflammation (e.g., rheumatoid arthritis), hypertension via macrophage infiltration, and cancer progression through impaired immune surveillance.
