Biological Activity

Description TLR4-IN-C34 (TLR4-C34, C34, TLR4-C34-IN) is a potent and selective antagonist of Toll-like receptor 4 (TLR4). TLR4-IN-C34 reduces systemic inflammation in mouse models of endotoxemia and necrotizing enterocolitis.

TLR4-IN-C34 inhibits TLR4 in vitro in macrophages and enterocytes via direct binding.[1]

In vivo

TLR4-IN-C34 can attenuate NEC severity and demonstrates a marked preservation of the intestinal mucosa.

  • Method

    IEC-6 cells and RAW 264.7 cells are treated withTLR4-IN-C34 at a concentration of 10 μM 30 min prior to treatment with LPS, and the extent of LPS signaling is determined by the degree of TNFα expression by qRT-PCR. In parallel, RAW 264.7 cells are transduced with an adenovirus expressing the NFκB-luciferase reporter gene, and then treated with LPS at 10 ng/ml after pre-treatment with TLR4-IN-C34 at 100 µM. The NFκB-luciferase activity is measured using the Luciferase Activity System.
