Description SB 4 is a potent and selective agonist of bone morphogenetic protein 4 (BMP4) signaling with EC50 of 74 nM. SB 4 enhances canonical BMP signaling and activates SMAD-1/5/9 phosphorylation.

BMP signaling agonist SB 4 induces phosphorylated SMAD-1/5/9 in a dose-dependent manner in PRECs cells under serum-starved conitions. SB 4 enhances the efficacy of signaling at each concentration of rhBMP4 tested. SB 4 increases BRE-luc expression 2-fold at 0.4 ng/ml of rhBMP4.[1]

    The BRE-Luc cells are stimulated with 2 ng/ml rhBMP4 for 1 h, after which the media are removed and replaced with fresh media containing either DMSO or 1 μm SB 4. p-SMAD-1/5/9 levels are then examined after 0, 5, 15, 30, 45, and 60 min of SB 4 treatment and compared with DMSO controls.
