Description QX77 is a novel activator of chaperone-mediated autophagy (CMA). QX77 induces the up-regulation of Rab11 expression and up-regulates LAMP2A expression.

QX77, an activator of CMA, eliminates the effects of milk fat globule-EGF factor 8 (MFG-E8) on TGF-β1-induced human pancreatic stellate cells (PSCs) activation. QX77 mitigates the suppressive effect of MFG-E8 on oxidative stress in activated PSCs.[2]

    Human pancreatic stellate cells (1×106/well) are treated with 5 ng/ml TGF-β1 with or without 20 ng/ml MFG-E8 for 24 h. 5 ng/mL QX77 is added simultaneously with 20 ng/mL MFG-E8 in TGF-β1-treated HPSCs. 24 h later, the cells are collected for various measurements. Western blot analyse the expression of GRP78, p-PERK, PERK, LAMP2A and MEF2D in the HPSCs .
