C45H38O18

Molecular Weight 866.77 CAS No. 37064-30-5
Solubility (25°C)* In vitro DMSO 100 mg/mL (115.37 mM)
In vivo (Add solvents to the product individually and in order)
Clear solution
5%DMSO 40%PEG300 5%Tween80 50%ddH2O
5.0mg/ml Taking the 1 mL working solution as an example, add 50 μL of 100 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to make it clear; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.
* <1 mg/ml means slightly soluble or insoluble.
* Please note that Selleck tests the solubility of all compounds in-house, and the actual solubility may differ slightly from published values. This is normal and is due to slight batch-to-batch variations.
* Room temperature shipping (Stability testing shows this product can be shipped without any cooling measures.)

Biological Activity

Description Procyanidin C1 is a polyphenolic component of grape seed extract, increases the healthspan and lifespan of mice through its action on senescent cells. PCC1 is a natural senotherapeutic agent with in vivo activity and a potential intervention to delay, alleviate or prevent age-related pathologies.
In vitro

Procyanidin C1 induces DNA damage, cell cycle arrest and enhanced the expression of checkpoint kinases. Procyanidin C1 decreases the level of Bcl-2 but increases the BAX, caspase 3 and 9 expression in cancer cells.[1]

In vivo

In rodent models, Procyanidin C1 depletes senescent cells in a treatment-damaged tumour microenvironment and enhances therapeutic efficacy when co-administered with chemotherapy. Intermittent administration of Procyanidin C1 to either irradiated, senescent cell-implanted or naturally aged old mice alleviates physical dysfunction and prolongs survival.[2]

Protocol (from reference)

Cell Assay:

[1]

  • Cell lines

    MDA-MB- 231 cell line, MCF-7 cell line

  • Concentrations

    48 h

  • Incubation Time

    6.25-100 mg/ml

  • Method

Overnight grown cells (5×103cells/well) are treated for 48 h with procyanidin C1 at a concentration ranging from 6.25 to 100 mg/ml in final volume of 150 ml. Cells without treatment served as control. After treatment, cells are used for MTT assay and cell cycle phase distribution analysis. Analysis of Bcl-2 or BAX levels is performed using an ELISA method.

Animal Study:

[2]

  • Animal Models

    C57BL/6J mice (males, 8–12 weeks)

  • Dosages

    20 mg/kg

  • Administration
