Biological Activity

Description Muscone (3-Methylcyclopentadecanone, Methylexaltone), a flavouring ingredient, is an organic compound that is the primary contributor to the odor of musk and also a potent anti-inflammatory agent. Muscone significantly downregulats the levels of LPS-induced inflammatory cytokines and inhibits NF-κB and NLRP3 inflammasome activation in BMDMs. Muscone remarkably decreases the levels of inflammatory cytokines (IL-1β, TNF-α and IL-6).

In vitro

Muscone induces CYP1A2 and CYP3A4 expression in liver cells in vitro and in vivo[1]. Muscone protects PC12 cells against glutamate-induced apoptosis by attenuating ROS generation and Ca2+ influx, via NR1 and CaMKII-depended ASK-1/JNK/p38 signaling pathways[2]. Muscone could promote bone marrow-derived mesenchymal stem cells (BMSCs) migration and proliferation[3].

In vivo

Muscone exhibits liver toxicity in Kunming mice at concentrations higher than 50 mg/kg[1]. It exerts potent neuroprotective activities on cerebral ischemia[2]. Muscone could accelerate the migration of BMSCs to the injured area in vivo[3].

    PC12 cells are seeded in 96-well culture plates at an approximate density of 1×105 cells per well and maintained in a CO2 incubator at 37 ℃ overnight. At the end of indicated treatment, MTT is added to the medium and incubated for another 4 h at 37℃. After the removal of the culture medium, the insoluble dark blue formazan crystal is dissolved with 150 μL DMSO. The absorbance (A) is measured at 490 nm with a microplate reader. Cell viability is expressed as a percentage with the control group as 100%.
