Biological Activity

Description L-citrulline is a substance called a non-essential amino acid that is used as a sports performance and cardiovascular health supplement.
In vitro L-Citrulline restores nitric oxide/reactive oxygen species balance and barrier function in high glucose-treated monolayers of human glomerular endothelial cells. Moreover, L-Citrulline also has the ability to establish an anti-inflammatory profile, characterized by increased IL-10 and reduced IL-1β and IL-12(p70) generation in the human proximal tubular cells[1].
In vivo Oral l-cit (precursor of l-arg) augments NO-dependent signaling, not only by means of increasing l-arg synthesis, but also by decreasing l-arg catabolism, as such increasing circulating l-arg levels. L-Citrulline exerts protective effects in kidneys of STZ-rats, and markedly reduces urinary albumin excretion, tubulo-interstitial fibrosis, and kidney hypertrophy, observed in untreated diabetic mice[1]. L-citrulline supplementation also strengthens vascular endothelium function and attenuates stress-induced cold hypersensitivity by improving blood circulation[2].

    Human glomerular endothelial cell are seeded at 1 × 105 cells per well in 24-well plates. Confluent quiescent cell monolayers are exposed to HG or proper iso-osmotic control for 24 h. L-Citrulline (1 mM) is applied 2 h prior to HG. Exposure is terminated by removal of the supernatant. Fresh basic CSC medium is replaced and cells incubated for additional 30 min. Supernatant is then removed, subsequently centrifuged and stored at −80°C for NO analysis.
