Biological Activity

Description JHU-083 (Ethyl 2-(2-Amino-4-methylpentanamido)-DON) is a novel prodrug of DON.JHU-083 selectively blocks glutaminase activity in brain CD11b+ cells and prevents depression-associated behaviors induced by chronic social defeat stress.Solutions are unstable and should be fresh-prepared.

JHU-083 selectively inhibits T-cell proliferation and decreases T-cell activation, with no effect on DCs.[2]

In vivo

In vivo, orally administered JHU-083 significantly decreases the experimental autoimmune encephalomyelitis(EAE) severity in both prevention and treatment paradigms and reverses EAE-induced cognitive impairment.[2]

    150,000 CD4+ T cells are plated with 20 μM eFlur450 cell proliferation dye, 2 μg/mL α-CD28, and ±JHU-083 in 100 μM complete RPMI. Every 24 hours, cells are treated with 20 μM media ±JHU-083.
