Biological Activity

Description DNPH1i (N6-benzyladenosine,BAPR) is a competitive inhibitor of adenosine deaminase(ADA) from L-1210 cells in axenic culture as well as a potent antiproliferative agent in vitro and in vivo.
In vitro

DNPH1i is the DNPH1 competitive inhibitor, that co-treatmented with hmdU selectively kills the HR-deficient cells, BRCA1-defective cells and PARPi-resistant BRCA1-mutant cell lines depleted of PARP1, sensitizing BRCA-deficient cells to PARPi or hmdU treatment.[2]

Protocol (from reference)

Cell Assay:

[2]

  • Cell lines

SUM149 BRCA1mut (parental) and WT (revertant) cell lines

  • Concentrations

    0.3 µM

  • Incubation Time

    8 days

  • Method

SUM149 BRCA1mut (parental) and WT (revertant) cell lines are either untreated or treated with hmdU in the absence or presence of DNPH1i (0.3 μM) for 8 days. Viability is determined using CellTiter-Glo.
