Biological Activity

Description Colivelin (CLN) is a brain-penetrant neuroprotective peptide with potent long-term capacity against deposition, neuronal apoptosis, and synaptic plasticity deficits in neurodegenerative disease. Colivelin is an activator of STAT3.

In vitro

Colivelin, a brain penetrant neuroprotective peptide, significantly increases the p-STAT3 protein levels in BV-2 cells.[2]

In vivo

Colivelin protects against ischemic brain injury and improves neurological outcomes, by elevating anti-apoptotic genes up-regulation and activates JAK/STAT3 signaling afetr ischemic stroke.[1]

  • Method

    BV-2 cells are maintained at 37℃ with 5% CO2. To build OGD models, BV-2 cells are washed with PBS for 3 times, and cultured in glucose-free DMEN and placed in a hypoxia incubation chamber filled with the gas mixture for 4 h at 37℃. The culture medium is then replaced by the normal medium containing 4.5 g/L glucose and cells are cultured under normoxic conditions for reoxygenation for 24 h. BV-2 cells are treated with 50 µg/ml rmMFG-E8 or/and Colivelin TFA, respectively. The expression changes of proteins (p-STAT3 and SOCS3) are analyzed by Western blot assay.
