Biological Activity

Description Calcium folinate (Leucovorin calcium) is a metabolically active, reduced form of folic acid that is rapidly absorbed and extensively metabolized in the liver to other folic acid derivatives.Solutions are unstable and should be fresh-prepared.
In vivo Calcium folinate (also known as Leucovorin) treatment has no effect on tumor growth. The combination of bortezomib and leucovorin is able to induce apoptosis and attenuate tumor growth in vivo[2].

  • Method

    The breast cells are grown in RPM11640 medium supplemented with 10% dialyzed fetal calf serum and 2 mM glutamine at an initial plating density of 500-1000 cells/plate. After 12 hr, the cells are treated for a 24-hr period with various MTX concentrations: 0.5, 1, 3, 10 μM. At the end of the 24-hr MTX exposure, the cells are washed two times with phosphate-buffered saline and placed in drug-free medium containing various concentrations of leucovorin. Colonies are allowed to grow for a lo-day period prior to staining and counting. Cloning efficiency is determined by comparing the number of colonies formed in MTX-treated cells to those of a control without MTX, but with exposure the corresponding leucovorin concentration.
