Description BC1618 is an orally active Fbxo48 inhibitor that stimulates adenosine monophosphate (AMP)-activated protein kinase (Ampk)-dependent signaling. This compound promotes mitochondrial fission, facilitates autophagy and improves hepatic insulin sensitivity.

This compound interrupts Fbxo48/pAmpkα interaction. It facilitates mitochondrial fission and autophagy.

In vivo

Consistent with augmenting Ampk activity, BC1618 promotes mitochondrial fission, facilitates autophagy and improves hepatic insulin sensitivity in high-fat-diet-induced obese mice.

  • Method

    293T cells transfected with Fbxo48-V5 or Fbxo30-V5 are treated with control (DMSO) or BC1618 (3 μM) for 30 min. Cells are then washed and equally fractioned into 10 aliquots and subjected to a range of increasing temperatures to promote protein denaturation. After a high-speed spin to precipitate heat-denatured proteins, the soluble protein fraction from each temperature is then immunoblotted for Fbxo48 or Fbxo30.
