Biological Activity

Description Amentoflavone (Didemethyl Ginkgetin) is a biflavonoid originally isolated from Selaginella with antibacterial, antioxidant, antiviral, antidiabetic, and neuroprotective activities.
In vitro Amentoflavone induces morphological changes and reduces the percentage of viable MCF-7 cells. Amentoflavone induces apoptosis and chromatin condensation, DNA strand breaks in MCF-7 cells. Amentoflavone is also reported to induce cell-cycle arrest at the sub-G1 phase, and apoptosis via mitochondria-emanated intrinsic pathways in SiHa and CaSki human cervical cancer cells[1].
In vivo In an animal model, amentoflavone is found to significantly inhibit B16F-10 melanoma-induced solid tumor development in C57BL/6 mice[1]. Amentoflavone can cross the blood-brain barrier. It effectively prevents pilocarpine-induced epilepsy in a mouse kindling model, suppresses NF-kB activation and expression, inhibits excessive discharge of hippocampal neurons resulting in a reduction in epileptic seizures, shortened attack time, and diminished loss and apoptosis of hippocampal neurons[2].

    Approximately 2×105 MCF-7 cells/well in 24-well plates are treated with 250 μM amentoflavone, or with vehicle (DMSO, 0.1% in culture media)-only and all cells are incubated for 6 to 72 h. For the examination of morphological changes, cells are photographed under a phase-contrast microscope.
