Biological Activity

Description Trolox is a cell-permeable, water-soluble derivative of vitamin E with potent antioxidant properties.
In vitro Trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-Carboxylic Acid) is a vitamin E analog and a direct scavenger of peroxyl and alkoxyl radicals. Trolox impairs PT4 cell survival, has minor effects on ROS levels and causes wide deregulation of cell cycle genes[1]. Trolox enhances As2O3-mediated apoptosis in APL, myeloma, and breast cancer cells. Trolox inhibits DNA damage formation induced by singlet oxygen in human lymphoblast WTK-1 cells and protects red blood cells during photodynamic treatment[2].
In vivo After cerebral ischemia reperfusion, Trolox prescription increases anti-apoptotic proteins and decreased proapoptotic proteins thus protects neurons of the hippocampus and caused improvement of memory[3].

  • Method

    NB4, IM9, and AsR2 cells are seeded at 1 × 105 cells/mL in 24-well plates. Cells are treated with various concentrations of As2O3 or doxorubicin, alone or in combination with 100 μM trolox for 6 days. Viable cells are counted by trypan blue exclusion on days 1, 3, and 6. All cells are maintained at a density lower than 1 × 106 cells/mL through dilution as required, and media ± treatment is replaced every third day. MCF-7, MDA-231, and T47D are seeded in 24-well plates at a density of 4000 cells/well. The next day, fresh media containing As2O3 ± trolox is added. On the days indicated, cells are fixed in 10% trichloroacetic acid and subsequently stained with sulforhodamine B (SRB). Bound SRB is solubilized in 10 mM unbuffered Tris (tris(hydroxymethyl)aminomethane), and optical density is measured at 570 nm in a microplate reader.
