Description Polyphyllin VI derived from Paris polyphylla possess anti-cancer activities.
In vitro

Polyphyllin VI (PPVI) induces activation of the NLRP3 inflammasome and exerts anti-proliferative effect via the induction of the ROS/NF-κB/NLRP3/GSDMD signal axis in non-small cell lung cancer (NSCLC).<sup><a class="sref"href="#s_ref">[1]</a></sup>

In vivo

Polyphyllin VI (PPVI), a main saponin isolated from TTM, activates the NLRP3 inflammasome in A549-bearing athymic nude mice.<sup><a class="sref"href="#s_ref">[1]</a></sup>

  • Method

    A549 or H1299 cells seeded onto 96-well cell plates at a density of 5×103 cells/well are treated with PPVI under the indicated concentrations for 24, 48, and 72 h. After treatment, 10 μL of MTT solution (5 mg/mL) is added to each well and continued to incubate for another 4 h. The medium is then removed and 150 L of DMSO is added to each well to dissolve the formed formazan in the cells. After shaking at low speed for 10 min at room temperature, the colorimetric reading of the solute mixture is then determined by spectrophotometer at OD 570 nm.
