Biological Activity

Description Picroside I (6'-Cinnamoylcatalpol), an iridoid glycoside, is a hepatoprotective agent which is reported to be antimicrobial and used against hepatitis B.
In vitro Picrosides I and II cause a concentration-dependent (> 0.1 μM) enhancement of basic fibroblast growth factor (bFGF, 2 ng/ml)-, staurosporine (10 nM)- and dibutyryl cyclic AMP (dbcAMP, 0.3 mM)-induced neurite outgrowth from PC12D cells. Picrosides activate MAP kinase-dependent signaling pathway[2].

  • Method

    For Western blot analysis, PC12D cells are plated at a cell density of 1×106 cells/35-mm culture dish, and cultured in the culture medium overnight. Cells are treated with or without picroside I or II 60 μM in the presence or absence of 7S NGF for 10 min. After the cells are washed with ice-cold PBS, they are lysed. Cell lysates are boiled for 5 min and subsequently sonicated for 3 min. Cell lysates are separated by SDS-polyacrylamide gel electrophoresis PAGE and transferred onto polyvinylidene difluoride membranes.
