Description Palmatine (Berbericinine, Burasaine), an isoquinoline alkaloid, has sedative, antidepressant, antioxidative, anti-ulcerative, antacid, anticancer, and anti-metastatic activities.

In vitro Palmatine plays an important role in osteoclast apoptosis via the NOS system in osteoclasts[1]. Palmatine has been reported to possess sedative and antioxidant activities. It has been also shown to be inhibitor of beta-site amyloid precursor protein-cleaving enzyme 1 (BACE 1), acetyl- and butyrylcholinesterases (AChE and BChE)[2].
In vivo Palmatine shows memory enhancing activity in mice probably by inhibiting brain acetylcholinesterase activity, through involvement of GABA-benzodiazepine pathway and due to its antioxidant activity[2].

  • Method

    Cell viability is measured using the cell counting kit-8 (CCK-8). The cells are seeded in a 96-well flat-bottomed microplate (5 × 103 cells/well) and cultured in 100 μL of growth medium at 37°C and 5% CO2 for 24 h or 5 days. The cell culture medium in each well is then replaced with 100 μL of cell growth medium containing palmatine at the following concentrations: 1, 5, 10, 40, and 100 µM. Additionally, L-NAME (10, 100, and 1000 μM) dissolved in PBS was added. After incubation for 24 h or 5 days at 37°C, the cells are washed with PBS 3 times. Then, 10 μL of CCK-8 dye and 100 μL of α-MEM cell culture medium are added to each well, and cells are incubated for another 1 h at 37°C. Cell viability (%) is calculated.
