Biological Activity

Description Gentisic acid (2,5-dihydroxybenzoic acid, 5-Hydroxysalicylic acid, Hydroquinonecarboxylic acid, DHB), an active metabolite of salicylic acid degradation, has a broad spectrum of biological activity, such as anti-inflammatory, antirheumatic and antioxidant properties.
In vitro

Gentisic acid (GA) has been shown to act as an antioxidant under different types of physical and chemical stimulus. Gentisic acid efficiently protects lipids, proteins, SOD enzyme and human erythrocytes on gamma radiation exposure. It has been found to scavenge physiologically relevant hydroxyl and peroxyl radicals in pulse radiolysis studies[3].

In vivo

Gentisic acid (GA) shows a potent protective effect against CP induced hepatotoxicity and genotoxicity. The protective effect of Gentisic acid can be attributed to its antioxidant properties, which enhance natural antioxidant enzymes and the cell membrane stabilizing property by inhibiting lipid peroxidation[2].

  • Method

    The Trypan Blue exclusion protocol is assessed in 24-well cell culture plates. In each well, 2.5×cells are seeded in 2 mL of complete culture medium with the following treatments: control (CO, 50 μL of culture medium, as negative control), cytotoxic agent doxorubicin (DXR, 10 μg/mL, as positive control), and Gentisic acid (0.08, 0.16, 0.8, 1.6, and 8 μg/mL). The cells are incubated for 24 h, trypsinized and then resuspended in 0.5 mL of culture medium. An aliquot of the cellular suspension (20 μL) is homogenized with Trypan Blue (0.2%) dye in a 1:1 ratio. The cells are quantified with a Neubauer chamber hemocytometer.
