Biological Activity

Description GC7 (N1-guanyl-1,7-diaminoheptane) Sulfate is an inhibitor of deoxyhypusine synthase (DHPS). GC7 inhibits Neuroblastoma (NB) cell proliferation in a dose-dependent manner, through induction of the cell cycle inhibitor p21 and reduction of total and phosphorylated Rb proteins.
Targets
DHPS [1] p21 [1]
In vitro

GC7 Sulfate, a deoxyhypusine synthase (DHPS) inhibitor, inhibits MYCN-amplified and chemotherapy-resistant NB cell proliferation, regulates several cell cycle controlling proteins and triggers arrest via the p21/Cdk4/Rb signaling axis, even in the presence of MYCN.[1]

Protocol (from reference)

Cell Assay:

[1]

  • Cell lines

    MYCN2 (±Dox) and BE(2)-C cells

  • Concentrations

    0.1-100 μM

  • Incubation Time

    72 h

  • Method

Cell viability was analyzed with the MTS assay kit. Briefly, after treatment with GC7 inhibitor for 72 h in 96-well plates, cells were directly incubated with the MTS dye reagent for 1 h at 37 ℃ in a 5 % CO2 atmosphere.
