Biological Activity

Description Bisdemethoxycurcumin (BDMC) is a natural demethoxy derivative of curcumin and possesses several biological activities, such as anti-inflammation and anti-cancer activities.
In vitro Bisdemethoxycurcumin (BDMC) is a demethoxy derivative of curcumin, which is a natural substance found in the turmeric root. BDMC inhibits the growth of gastric cancer SGC 7901 cells in a dose‑dependent manner. BDMC arrests the cell growth in G1 phase and thus, cells could not reach S phase and complete the cycle[2].
In vivo BDMC may promote apoptosis in tumors via mitochondrial modulation. BDMC inhibits the tumor growth and caused an increase in body weight of gastric adenoadenocarcinoma xenograft model[2].

Protocol (from reference)

Cell Assay:

[1]

  • Cell lines

The NSCLC cell lines A549, H460 and SPC-A-1

  • Concentrations

    0-100 μM

  • Incubation Time

    72 h

  • Method

The cells are grown at 37˚C in a humidified atmosphere of 5% CO2. To evaluate the hypomethylation effect of bisdemethoxycurcumin, DAC is used as positive control. Curcumin, DMC, BDMC and DAC are prepared as stock solutions (20 mM) in dimethyl sulfoxide (DMSO) and diluted in sterile-filtered media before use. Cells (1×105) are seeded into 6-well culture plates. After 24 h of culture, the cells are treated with curcumin, DMC, BDMC (0-100 μM) or DAC (20 μM) for 72 h.

Animal Study:

[2]

  • Animal Models

human gastric adenoadenocarcinoma xenograft model (nude mice)

  • Dosages

    100 mg/kg/day

  • Administration

    s.c.
