Biological Activity

Description Alpha-mangostin is the main xanthone purified from mangosteen and has health promoting benefits including anti-bacterial, anti-inflammatory, anti-oxidant, anti-cancer and cardioprotective activities. alpha-Mangostin (α-Mangostin) is a dietary xanthone with broad biological activities, such as antioxidant, anti-allergic, antiviral, antibacterial, anti-inflammatory and anticancer effects. It is an inhibitor of mutant IDH1 (IDH1-R132H) with a Ki of 2.85 μM. Alpha-mangostin is also an agonist of human STING.
In vitro α-mangostin shows both fast-binding and slow-binding inhibitions to FAS in vitro. It inhibits FAS overall reaction with a half-inhibitory concentration (IC50) value of 5.54 μM non-competitively with respect to NADPH, and partially competitively against both substrates acetyl-CoA and malonyl-CoA. α-Mangostin could reduce cell viability and induce apoptosis in human breast cancer cells[1]. Alpha-mangostin significantly inhibits light-induced degeneration of photoreceptors and 200 μM H2O2-induced apoptosis of RPE cells. Alpha-mangostin can activate the expression of PKC-δ and down-regulate the expression of mitogen-activated protein kinases (MAPKs), including ERK1/2, JNK, P38[2].
In vivo Alpha-mangostin stimulation results in an increase of superoxide dismutase (SOD) activity, glutathione peroxidase (GPX) activity and glutathione (GSH) content both in vivo and vitro. It suppresses light-induced retina dysfunction. Alpha-mangostin has protective effect on kidney, heart and brain tissues against oxidative stress[2].

  • Method

    MCF-7 and MDA-MB-231 cells are treated with 0, 1, 2, 3, 4, 6, 8, and 10 μM α-mangostin for 24 h and 48 h. Cell viability is then determined by the CCK-8 assay.
