Description

Procyanidin B1, a natural polyphenolic compound that has antioxidant properties, inhibits Kv10.1 channel and suppresses the evolution of hepatoma.

In vitro

Procyanidin B1 is a polyphenolic flavonoid isolated from fruits and fruit juices, which inhibits LPS-induced TNF-α production, and expression of MD-2, TRAF6, NF-κB mRNA, phosphorylated p38 MAPK and NF-κB protein in THP1 cells.^[1]

In vivo

Procyanidin B1 selectively inhibits the Kv10.1 channel in a concentration-dependent manner, which significantly suppress the growth of the tumor (HepG2) with no side effect on the normal metabolismof the mice.^[2]

Total RNA is extracted from THP1 cells using the RNAiso Plus kit. Isolated total RNA is reverse transcribed using an RT master mix kit for 15 min at 37℃ and 5 s at 85℃. cDNAs are amplified using a 7500 Real-Time PCR System with sequence-specific primers for MD-2, TLR4, TRAF6, and NF-kB designed and synthesized by Takara Biotechnology.

Method
