Biological Description

Specificity Phospho-PKCθ/PRKCQ (Thr538) Antibody [D2M12] detects endogenous levels of total PKCθ/PRKCQ protein only when it is phosphorylated at Thr538.
Background Phospho-PKCθ/PRKCQ (Thr538) is the activated form of protein kinase C theta (PKCθ), a key enzyme in T cells. PKCθ belongs to the novel PKC family and helps T cells respond to signals from their receptor. It has special regions called C1 domains that bind a molecule called diacylglycerol (DAG), which helps PKCθ move to the cell membrane when T cells are activated. The phosphorylation (a chemical modification) at threonine 538 (Thr538) in the activation loop of PKCθ is essential because it turns the enzyme "on," allowing it to perform its function. Without this phosphorylation, PKCθ cannot carry out its kinase activity, even though it can still move to the membrane. This shows that membrane location and enzyme activation are controlled separately. When active, PKCθ helps activate important pathways that control T cell growth, division, and immune responses. It also plays a role in diseases like autoimmune disorders when its activity is not properly regulated.
