Biological Description

Specificity PDGFR α Rabbit mAb recognizes endogenous levels of total PDGFR α protein.
Background PDGFRα (Platelet-derived growth factor receptor alpha) is a type III receptor tyrosine kinase characterized by five extracellular immunoglobulin (Ig)-like domains, a single transmembrane domain, and an intracellular region containing a split kinase domain with a unique inserted sequence, a juxtamembrane domain, and a variable C-terminal tail. Activated by ligand-induced dimerization and autophosphorylation, PDGFRα forms homo- or heterodimers (e.g., with PDGFRβ) upon binding PDGF ligands, excluding PDGF-D. PDGFRα is predominantly expressed in cranial neural crest (CNC)-derived mesenchyme and is essential for embryonic development, particularly craniofacial and tooth morphogenesis. It regulates cell proliferation, survival, and migration in a paracrine manner, with PDGFRα expressed in odontogenic mesenchyme and its ligand PDGF-A in dental epithelium throughout all stages of tooth development. Loss of PDGFRα function results in craniofacial defects, mid-facial cleft, and embryonic lethality, highlighting its critical role in CNC-derived tissue development.
