Specificity P2X7 Receptor Antibody [L17L11] detects endogenous levels of total P2X7 receptor protein.
Background The P2X7 receptor (P2X7R) is a unique member of the ATP-gated cation channel P2X family, defined by its requirement for unusually high extracellular ATP concentrations for activation. Structurally, each P2X7R subunit is a 595–amino acid protein encoded by the P2RX7 gene on chromosome 12q24.31, comprising two transmembrane helices, a large glycosylated extracellular loop harboring ATP-binding sites, and intracellular N- and C-terminal domains, with the extended C-terminus containing unique regulatory motifs. Three subunits assemble into a homo-trimeric receptor, though occasional hetero-trimerization with P2X4 and other P2X subunits occurs. P2X7R is broadly expressed in immune cells, glial cells, and various peripheral tissues, with debated neuronal expression. Functionally, it mediates rapid Na⁺ and Ca²⁺ influx and K⁺ efflux upon ATP binding; with prolonged stimulation, it can form large pores permeable to organic cations, triggering inflammatory signaling, cytokine release, and cell death. This receptor plays critical roles in immunity, inflammation, neurodegeneration, cancer progression, and psychiatric disorders, making it a key therapeutic target.
