Clear solution
5%DMSO 40%PEG300 5%Tween80 50%ddH2O
2.5mg/ml Taking the 1 mL working solution as an example, add 50 μL of 50 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.
Clear solution
5% DMSO 95% Corn oil
0.6mg/ml Taking the 1 mL working solution as an example, add 50 μL of 12 mg/ml clear DMSO stock solution to 950 μL of corn oil and mix evenly. The mixed solution should be used immediately for optimal results.
* <1 mg/ml means slightly soluble or insoluble.
* Please note that Selleck tests the solubility of all compounds in-house, and the actual solubility may differ slightly from published values. This is normal and is due to slight batch-to-batch variations.
* Room temperature shipping (Stability testing shows this product can be shipped without any cooling measures.)

Biological Activity

Description MF-094, a potent and selective USP30 inhibitor (IC50=0.12 μM), accelerates diabetic wound healing by inhibiting the NLRP3 inflammasome.
Targets
USP30 [1]
0.12 μM
In vitro

MF-094 restores viability and migration of AGE-treated HSF2 cells. MF-094 treatment decreases protein levels of NLRP3 and its downstream target caspase-1 p20.[2]

In vivo

MF-094 treatment facilitates wound healing in diabetic rats.[2]

Protocol (from reference)

Cell Assay:

[2]

  • Cell lines

    HSF2 human skin fibroblasts

  • Concentrations

    100 nM

  • Incubation Time

    24 h

  • Method

HSF2 cells are treated with USP30 inhibitor MF-094 (100 nM) or transduced with shUSP30 lentivirus vector for 24 h, following 200 μg/ml AGE treatment for 24 h. After treatment, HSF2 cells (5 × 105 cells/well) are plated in 96- well plates and cultured for 0, 12, 24, and 48 h. Then, 10 ml CCK-8 solution is added to cells for cell viability assay. The protein levels of NLRP3 and P20 are determined by Western blot.

Animal Study:

[2]

  • Animal Models

    Streptozotocin (STZ) induced diabetic rats

  • Dosages

    50 mg/kg

  • Administration
