Biological Description

Specificity MBD2 + MBD3 Mouse mAb detects endogenous levels of total MBD2 + MBD3 protein.
Background MBD2 + MBD3 are closely related members of the methyl-CpG-binding domain (MBD) protein family, which function as epigenetic readers that influence chromatin structure and gene expression. Both proteins are expressed in various tissues, with MBD3 being essential for early embryonic development, while MBD2 is dispensable but influences processes such as maternal behavior. Structurally, they share a conserved MBD and a coiled-coil domain, but MBD2 uniquely contains an N-terminal glycine-arginine-rich region and a transcriptional repression domain (TRD), enabling it to bind methylated DNA and recruit the Mi-2/NuRD complex to silence transcription. In contrast, MBD3 lacks key residues in the MBD necessary for methylated DNA binding but remains a core component of the NuRD complex, where it regulates chromatin remodeling and differentiation. Despite their high sequence similarity and shared association with NuRD, MBD2 and MBD3 perform distinct, non-redundant roles in development and gene regulation, with MBD2 acting more directly in methylation-dependent repression and MBD3 playing a central role in lineage commitment and pluripotency control.
