Biological Description

Specificity LATS1 Antibody [A5C10] detects endogenous levels of total LATS1 protein.
Background LATS1 (Large Tumor Suppressor Kinase 1) is a serine/threonine kinase of the AGC family and a core component of the Hippo signaling pathway. Structurally, it contains an N-terminal regulatory domain (NTR), a catalytic kinase domain with a unique insert between subdomains VII and VIII, and two conserved phosphorylation sites: the activation segment (AS; Ser909) and hydrophobic motif (HM; Thr1079), essential for its activity. LATS1 is widely expressed in mammalian tissues, where it acts as a tumor suppressor by phosphorylating YAP/TAZ to inhibit their nuclear localization and transcriptional co-activation, thereby regulating cell proliferation, apoptosis, and differentiation. Its regulation involves multiple phosphorylation events by upstream kinases (MST1/2, MAP4Ks, CDK1/cyclin B, NUAK1), adaptor proteins (MOB1, SAV1, angiomotins), and control of protein stability by HSP90, ROS-PKCδ signaling, and ubiquitin ligases, highlighting its central role in maintaining tissue homeostasis and preventing tumorigenesis.
