Biological Activity

Description Kojic acid is a fungal metabolite that inhibits tyrosinase with an IC50 value of 30.6 µM for mushroom tyrosinase. It is a synthetic intermediate used for production of food additives.

In vitro Kojic acid (KA) acts as a good chelator of transition metal ions such as Cu2+ and Fe3+ and a scavenger of free radicals. Kojic acid shows a competitive inhibitory effect on the monophenolase activity and a mixed inhibitory effect on the diphenolase activity of mushroom tyrosinase[1]. Kojic acid could highly protect Chinese hamster ovary (CHO) cells against ionizing radiation with low toxicity[3].
In vivo Kojic acid exerts its radioprotective effects on lethal or sub-lethal irradiation induced damages in mice[3].

    CHO cells are incubated in RPMI 1640 medium containing 10% (v/v) fetal bovine serum, and maintained at 37°C in a 5% CO2 incubator. For cytotoxicity assay, CHO cells are seeded in 96-well plates at a density of 2500 cells per well in 180 μl medium. Then, 20 μl of Kojic acid stock solution are added to each well to make sure Kojic acid at the final concentrations of 0, 0.1, 1, 10, 100, 1000, 1500, 2500 μg/ml. After cultured for 72 h, each well of the cells is added with 20 μl MTT (5 mg/mL), which is dissolved in sterile Phosphate Buffered Saline (pH 7.4) and incubated for 4 h at 37°C. Then thesupernatant is discarded and 200 μl DMSO is added to each well. The mixture is sufficiently oscillated in the dark place for 15 min. The optical density values of wells are measured at 492 nm.
