Biological Activity

Description Isoleucine (Ile) is a non-polar, uncharged (at physiological pH), aliphatic amino acid that is used in the biosynthesis of proteins.
In vitro Isoleucine plays a major role in enhancing glucose consumption and utilization by up-regulating intestinal and muscular glucose transporters[1]. L-isoleucine could induce β-defensins’expressions in porcine intestinal epithelial cells[2].
In vivo Isoleucine is beneficial for mammary health, milk quality and embryo growth, enhances intestinal development, intestinal amino acid transportation and mucin production and participates in up-regulating innate and adaptive immune responses. Studies in mice have indicated that diet-induced obesity mice in an isoleucine treatment (final concentration of 2.5% isoleucine in drinking water) had almost 6% lower body weight gain and 49% less epididymal white adipose tissue mass compared with the control treatment, with higher levels of hepatic protein. Its deprivation induces fat mass loss in mice. Isoleucine could stimulate glucose uptake and participated in plasma glucose uptake in the rat[1].

  • Method

    After cells are grown to 80 % confluence, they are starved for 12 h in serum and antibiotic-free DMEM/F12 medium. Following starvation, the cells are cultured for 12 h in the presence of 0, 25, 50, 100, 250 and 500 μg/mL L-isoleucine or 0, 25, 50, 100, 250 and 500 μmol/mL zinc sulfate that were used to increase the concentration of Zn2+ in the medium. Then, the cells are collected to examine the mRNA and protein expressions of pBD-1, 2 and 3, respectively.
