Biological Activity

Description GsMTx4 (Grammostola spatulata mechanotoxin 4, M-theraphotoxin-Gr1a, M-TRTX-Gr1a) is a spider venom peptide that inhibits cationic mechanosensitive channels (MSCs).
Targets
MSCs [1]
In vitro

Pretreatment of GsMTx4 and BAPTA-AM suppress the Yoda1-induced accumulation of HIF-1alpha, NFATC1, and NFATC3 proteins and also suppress Yoda1-induced VEGF-A expression at the mRNA and protein levels.[2]

Density 95.0~105.0%

Protocol (from reference)

Cell Assay:

[3]

  • Cell lines

    BMDMs, RAW264.7 cells

  • Concentrations

    5, 10 µM

  • Incubation Time

    30 min

  • Method

To study the effect of mechanical stretching, BMDMs or RAW264.7 cells were treated with Yoda1, with or without pretreatment with inhibitors for 30 min. The inhibitors included cyclosporin A, FK506, echinomycin, GsMTx4, BAPTA-AM and RN-1734. In some experiments, BMDMs were treated with GSK1016790A.
