Background GOPC (Golgi-associated PDZ and coiled-coil motif-containing protein), also known as FIG or PIST, is a pivotal scaffolding protein localized primarily to the trans-Golgi network, characterized by two N-terminal coiled-coil domains and a C-terminal PDZ domain. Its main functional role centers on mediating vesicular trafficking and regulating the sorting and surface expression of various membrane proteins, including ion channels, receptors, and adhesion molecules. GOPC interacts with golgin-160 and the Q-SNARE protein syntaxin 6 through its coiled-coil domains, facilitating cargo transport in secretory and endocytic pathways. The PDZ domain specifically binds to the C-termini of target proteins such as Frizzled receptors, neuroligins, and CFTR, thus influencing processes like cell signaling, tight junction integrity, and receptor recycling.
