Biological Description

Specificity Fragilis Antibody [E3D3] detects endogenous levels of total Fragilis protein.
Background Fragilis, also known as Ifitm3 in mouse and a member of the interferon-inducible Fragilis/IFITM family, is a small type I membrane protein characterized by two hydrophobic transmembrane domains and short luminal and cytosolic regions, with expression detected in extraembryonic ectoderm and epiblast and robustly upregulated by interferon signaling in various tissues. The Fragilis family consists of a cluster of interferon-inducible transmembrane genes located on mouse chromosome 7—including Fragilis, Fragilis2, Fragilis3, Fragilis4, and Fragilis5—with conserved human orthologs present on the syntenic region of chromosome 11; all family members display a predicted membrane topology comprising a luminal N-terminus, a conserved intracellular loop, and a C-terminal cytosolic tail, a configuration linked to functions in membrane organization and signaling. Fragilis represents one of the earliest markers of primordial germ cell competence: its expression expands within the proximal epiblast before germ cell fate specification, and Fragilis-positive cells include both future germ cell precursors and neighboring somatic cells with germ cell potential. Single-cell analyses and in situ hybridization studies reveal that transcripts for Fragilis, Fragilis2, and Fragilis3 are highly enriched in nascent primordial germ cells and remain predominant in gonadal germ cells, establishing Fragilis family expression as a marker of the germ cell lineage from initial specification through colonization of the gonads. The interferon responsiveness of the Fragilis locus, together with its developmental expression pattern, positions Fragilis at the interface of cytokine signaling and germline specification, in line with models proposing that interferon-regulated pathways participate in establishing germ cell competence and identity during early development.
