Flotillin-1 (J14F12) Rabbit mAb detects endogenous levels of total Flotillin-1 protein.

Background

Flotillin 1 is a conserved membrane-associated protein of the SPFH (Stomatin, Prohibitin, Flotillin, HflK/C) superfamily that functions as a core scaffolding component of cholesterol-rich lipid rafts, where it forms hetero‑oligomeric complexes with Flotillin 2 to organize membrane architecture and signaling platforms. It contains an N‑terminal SPFH domain that anchors to the inner plasma membrane and mediates oligomerization, and a C‑terminal flotillin domain that stabilizes higher‑order structures through coiled‑coil and β‑strand interactions. Flotillin 1 regulates clathrin‑independent endocytosis, receptor clustering, insulin and immune receptor signaling, cytoskeletal organization, vesicle trafficking, and lateral membrane compartmentalization, also localizing to endosomes, exosomes, lysosomes, and sometimes the nucleus. By inducing membrane curvature and providing stable nanodomains, it coordinates pathways governing cell adhesion, motility, and signal transduction. Dysregulated Flotillin 1 is associated with cancer progression by promoting metastasis through modulation of oncogenic signaling, and with neurodegenerative and metabolic disorders through disturbances in membrane organization and intracellular trafficking.
