Biological Activity

Description FIDAS-3 effectively competes against S-adenosylmethionine (SAM) for methionine sadenosyltransferase 2A (MAT2A) binding . FIDAS-3 has anticancer activities by inhibiting the expression of oncogenes. FIDAS-3 is a stilbene derivative and is a potent Wnt inhibitor with an IC50 of 4.9 μM for MAT2A.
Targets
hMat2A [1]
4.9 μM
In vitro

FIDAS-3 inhibits Methionine S-adenosyltransferase 2A (MAT2A) activity in S-adenosylmethionine (SAM) synthesis, leading to the inhibition of CRC cell growth.[1]

In vivo

FIDAS-3 represses the growth of human CRC xenografts in athymic nude mice with no notable effects on body weight.FIDAS-3 represses the growth of human CRC xenografts in athymic nude mice with no notable effects on body weight. [1]

Protocol (from reference)

Cell Assay:

[1]

  • Cell lines

    LS174T cells

  • Concentrations

    3-10 μM

  • Incubation Time

    36 h

  • Method

For cell based assay(Western blotting ; Cell proliferatio; shRNA assay), LS174T cells are cultured in RPMI1640 medium containing 5% FBS. Cells are treated with FIDAS agents for 36 h and then harvested and weighed. Perchloric acid (0.4 M) is added to cell pellet (100 μL/30mg) for deproteinization.

Animal Study:

[1]

  • Animal Models

Athymic nude mice bearing with HT29 CRC cells

  • Dosages

    20 mg/kg

  • Administration

    o.g.
