Biological Description

Specificity Desmoglein 2/DSG2 Antibody (Rabbit mAb) [M16H3] detects endogenous levels of total Desmoglein 2/DSG2 proteins.
Background Desmoglein 2 (Dsg2) is a calcium-dependent desmosomal cadherin that, together with its binding partner desmocollin 2, forms the adhesive core of desmosomes, the intercellular junctions that anchor the intermediate filament cytoskeleton at sites of cell-cell contact in epithelial tissues and cardiac muscle. The ectodomain of Dsg2 mediates trans-interactions with desmosomal cadherins on the surface of an adjacent cell through exchange of N-terminal beta-strands, and solution-based structural analysis shows that this ectodomain remains flexible even in its calcium-bound, adhesion-competent state and is shorter on average than classical cadherin structures, a flexibility that distinguishes desmosomal cadherin architecture from the more rigid adherens junction cadherins and helps explain the distinct mechanical properties of desmosomes compared with adherens junctions. Beyond its adhesive role, Dsg2 also engages classical cadherins including E-cadherin and N-cadherin in calcium-dependent interactions, providing molecular cross-talk between desmosomes and adherens junctions in tissues such as cardiac myocytes and intestinal epithelium where both junction types coexist. In intestinal enterocytes, where Dsg2 is the only desmoglein isoform expressed, an antibody that blocks Dsg2-specific binding events at the cell border activates p38 mitogen-activated protein kinase signaling without reducing cell-cell cohesion, and cells genetically deficient in Dsg2 show reduced p38MAPK activity together with impaired epithelial barrier integrity and delayed barrier reformation after disruption. Substituting the related desmosomal cadherin desmocollin 2 into Dsg2-deficient cells fails to restore p38MAPK activity, demonstrating that Dsg2 itself, rather than desmosomal adhesion in general, is specifically required to activate this signaling pathway, establishing Dsg2 as a signaling receptor for epithelial barrier maintenance in addition to its structural adhesive function.
