Biological Description

Specificity DC-SIGN + DC-SIGNR Antibody (Rabbit mAb) [C17K8] detects endogenous levels of total DC-SIGN and DC-SIGNR protein.
Background DC-SIGN (CD209) and DC-SIGNR (L-SIGN, CD209L) are closely related transmembrane C-type lectins built from an N-terminal cytoplasmic tail, a tandem-repeat neck region and a C-terminal carbohydrate recognition domain, with the neck domain driving oligomerization into tetramers that allow multivalent, high-avidity binding to glycan ligands. DC-SIGN is expressed on dendritic cells and recognizes high-mannose glycoproteins displayed on the envelopes of a wide range of microorganisms, functioning simultaneously as a cell-adhesion receptor, through binding ICAM-3 and ICAM-2, and as a pathogen-recognition receptor that captures viral, bacterial and parasitic ligands. DC-SIGNR shares this carbohydrate-recognition architecture but is expressed on liver and lymph node sinusoidal endothelial cells rather than on dendritic cells, giving the two receptors overlapping ligand specificity but distinct tissue distribution and physiological roles. Captured viral particles bound by DC-SIGN or DC-SIGNR are internalized into nonlysosomal intracellular compartments rather than degraded, and this protects the virus from destruction during transport, allowing subsequent transfer of intact virions to permissive target cells expressing the bona fide entry receptor, a process termed trans-infection. Hepatitis C virus particles captured by DC-SIGN on dendritic cells or by DC-SIGNR on liver sinusoidal endothelial cells are transferred by this mechanism to hepatocytes, and this receptor-mediated capture and relay function extends to HIV-1, which DC-SIGN-bearing dendritic cells transport from sites of mucosal exposure to lymphoid organs where the virus is transmitted to CD4 T cells. Because this capture-and-transfer mechanism protects pathogens from immune clearance rather than eliminating them, DC-SIGN and DC-SIGNR represent points of vulnerability that several viruses exploit to establish systemic infection.
