Biological Description

Specificity Cleaved Caspase-3 Antibody [G2J3] detects endogenous levels of activated caspase-3.
Background Cleaved caspase-3 is the active form of caspase-3, a key executioner protease in apoptosis and a member of the Cysteine-ASPartic proteASES family (cysteine proteases). It is generated by proteolytic cleavage of procaspase-3 at specific aspartate residues, producing large (p17) and small (p12) subunits that assemble into a functional heterotetramer. This active enzyme is widely expressed across tissues but shows increased levels in pathological conditions such as cancer, where its expression often marks apoptotic activity. Functionally, cleaved caspase-3 orchestrates the execution phase of apoptosis by cleaving crucial substrates, including ICAD, PARP, and cytoskeletal proteins, thereby driving DNA fragmentation, chromatin condensation, and the characteristic morphological changes of apoptotic cells. Beyond apoptosis, emerging evidence also implicates cleaved caspase-3 in non-apoptotic roles, such as transcriptional regulation and angiogenesis, highlighting its dual role in cell death and tumor progression.
