Biological Activity

Description Auxinole is a potent auxin antagonist that binds to TIR1 and blocks the formation of the TIR1-IAA-Aux/IAA complex. Auxinole is an OsTIR1 inhibitor that suppresses leaky degradation of degron-fused proteins.
In vitro

Auxinole causes reduction in IAA-triggered depolarization in root hair cells. Auxinole (20 µM) also represses the transient increase in [Ca2+]cyt completely, and blocks Ca2+ response.[4]

    Sterile grown seedlings are exposed to standard bath solution (0.1 mM KCl, 1 mM CaCl2, 5 mM MES/BTP pH 5.5; in the case of La3+-containing bath solutions ionic strength is kept constant and the sum of La3+ and Ca2+ concentrations is 1 mM). The response of the membrane potential of root epidermal cells to apoplastic and cytosolic auxin application is measured by sharp single- and double-barreled microelectrodes impaled through the tip of bulging root-hair cells. Ion fluxes across the plasma membrane of root epidermal cells are monitored using non-invasive ion-selective scanning microelectrodes.
