Specificity Anti-TIMP3 Rabbit Antibody [M11A20] detects endogenous levels of total TIMP3 protein.
Background Tissue Inhibitor of Metalloproteinase 3 (TIMP3) is a unique member of the TIMP family that binds tightly to the extracellular matrix (ECM) and inhibits a broad spectrum of enzymes, including matrix metalloproteinases (MMPs), a disintegrin and metalloproteinases (ADAMs), and ADAM with thrombospondin motifs (ADAMTSs). TIMP3 consists of an N-terminal domain responsible for its inhibitory activity and a C-terminal domain that influences its binding to ECM components and specific metalloproteinases. TIMP3 regulates ECM turnover, inflammation, angiogenesis, and cell apoptosis by inhibiting proteolytic enzymes that degrade ECM and modulating signaling molecules such as TNF-α through ADAM17 inhibition. Its inhibitory action on ADAM17 and ADAMTS4 also links TIMP3 to controlling processes like tumor progression and osteoarthritis. Regulation of TIMP3 expression is complex, involving transcription factors such as Sp1, AP1, and Smads, and post-transcriptional regulation by numerous microRNAs targeting its long 3’ untranslated region (UTR). Growth factors like TGF-β1 stimulate TIMP3 expression via ERK1/2 and Smad pathways, whereas microRNAs such as miR-205 negatively regulate its levels with implications in cardiovascular and inflammatory diseases. TIMP3 mutations or dysregulation contribute to pathological fibrosis, cancer metastasis, and other ECM-related disorders.
