Biological Description

Specificity Anti-RCC1 Rabbit Antibody [F6F22] detects endogenous levels of total RCC1 protein.
Background RCC1 (Regulator of Chromosome Condensation 1) is a 45-kDa nuclear protein and the sole guanine nucleotide exchange factor (GEF) for the small GTPase Ran, essential for cell cycle progression and nuclear transport. Structurally, RCC1 consists of a short N-terminal domain (NTD) containing a lysine-rich nuclear localization signal (NLS) and a major catalytic domain that adopts a seven-bladed β-propeller fold, enabling direct interaction with histones H2A/H2B and nucleosomal DNA. It is ubiquitously expressed in proliferating cells, where it associates with chromatin throughout the cell cycle. Functionally, RCC1 maintains the Ran-GTP gradient by catalyzing the exchange of GDP for GTP on Ran, thereby regulating nucleocytoplasmic transport, mitotic spindle assembly, nuclear envelope reformation, and chromosome condensation. Proper nuclear and chromatin localization of RCC1 is critical, as it ensures asymmetric Ran-GTP distribution, which underlies its essential role in nuclear organization and cell division.
