Biological Description

Specificity Anti-IFI16 Rabbit Antibody [F5E19] detects endogenous levels of total IFI16 protein.
Background IFI16 is a prominent sensor of nuclear pathogenic DNA, initiating innate immune signaling and suppressing viral transcription. It is a nuclear protein belonging to the interferon-inducible HIN-200 family, characterized by an N-terminal PYRIN domain, which is involved in apoptosis, inflammation, and immune responses, and two C-terminal HIN domains, each comprising tightly packed OB-fold subdomains. It is widely expressed in human endothelial, epithelial, and hematopoietic cells, with reduced expression often linked to cancers such as pancreatic, prostate, and breast tumors, suggesting a tumor suppressive role. Structurally, the HIN-A domain of IFI16 binds to the basic C terminus of p53, while the HIN-B domain interacts with its core DNA-binding region, together enhancing p53-DNA complex formation and transcriptional activation. Functionally, IFI16 regulates transcription through protein-protein interactions with p53, pRB, E2F1, and BRCA1, modulates p53 stability, and promotes apoptosis in response to DNA damage, thereby playing a central role in tumor suppression, innate immune signaling, and cellular stress responses.
