Specificity Anti-CAP1 Rabbit Antibody [C11L24] recognizes total endogenous levels of CAP1 protein.
Background Cyclase-associated proteins (CAPs) are evolutionarily conserved actin-binding proteins found in all eukaryotes. They regulate actin dynamics by engaging in distinct interactions: the N-terminal domain associates with the adenylyl cyclase complex, while the C-terminal region binds directly to monomeric actin. Loss of CAP function disrupts cytoskeletal organization, underscoring its role in actin regulation. Among them, adenylyl cyclase-associated protein 1 (CAP1) is a ubiquitously expressed isoform that modulates cell adhesion through interactions with talin1 and FAK (Focal Adhesion Kinase). Elevated CAP1 expression has been linked to cancer progression and metastasis, including in hepatocellular carcinoma, esophageal squamous cell carcinoma, and epithelial ovarian cancer, where it correlates with proliferation, histological grade, and prognosis. CAP1 also promotes cell migration in mammalian systems, supporting its role in cancer cell motility and invasiveness. In breast cancer, CAP1 influences proliferation via regulation of ERK (Extracellular signal-Regulated Kinase) signaling. Notably, CAP1 depletion alters both FAK and ERK activities, producing opposite adhesion and proliferation phenotypes in metastatic versus non-metastatic breast cancer cells.
